Did you know that there are direct and indirect methods of measuring Hemoglobin A1c?

Many HbA1c methods are indirect measurements, meaning that the HbA1c concentration and the concentration of hemoglobin are both measured. The percent HbA1c in the sample is then calculated as follows: % HbA1c = ([HbA1c] / [Total Hemoglobin]) x 100. Both the concentration of hemoglobin A1c specifically, and the concentration of total hemoglobin, are measured. The ratio is reported as a percent hemoglobin A1c.

The direct HbA1c method utilizes the interaction of antigen and antibody to directly determine the HbA1c concentration in whole blood. This method is possible due to MedTest’s Pointe Scientific brand’s proprietary antibody whose epitope on the N-terminal of the B-chains does not require denaturation of the hemoglobin molecule in order to be available to the antigen binding fragments (Fab). These reagents have the highest degree of antibody specificity, minimizing the cross-reactivity from hemoglobin variants (HbA2, HbC, HbD, HbE and HbS).
